A 2025 systematic review in Molecular Nutrition and Food Research examined edible medicinal fungi, Lion's Mane among them, and mapped out the mechanisms researchers are investigating. The proposed pathways included regulation of the HPA axis, modulation of serotonin and dopamine signalling, and a reduction in neuroinflammation.

What the Human Trials Have Shown

The most cited human study is small but notable. In 2010, Nagano and colleagues ran a randomised, placebo-controlled trial in which thirty women took either Lion's Mane or placebo for four weeks. The Lion's Mane group showed significantly lower scores on a validated depression scale after the four weeks, and lower scores on several markers of the vague, hard-to-name complaints that so often accompany chronic stress, including irritability and anxiety.

More recently, a 2023 pilot trial published in Nutrients gave healthy adults Lion's Mane for 28 days. The researchers observed a trend towards reduced subjective stress, though it fell just short of statistical significance, alongside faster performance on a cognitive processing task after a single dose.

In 2025, a UK study at the University of Surrey gave healthy young adults a single 3g dose of 10:1 Lion's Mane fruiting body extract and found no significant overall improvement in mood or global cognition, with only one specific task showing a benefit.

Two things are worth drawing from that. First, a single dose is not the same as sustained daily use, and the mechanisms most relevant to burnout, nerve growth factor and stress-axis regulation, are thought to build over weeks, not minutes. Second, the human evidence base as a whole is still small, and larger, longer, Lion's Mane-specific trials on stress and fatigue are the obvious next step. A 2025 systematic review in Frontiers in Nutrition reached a similar conclusion: promising signals on mood, anxiety and related symptoms, tempered by a call for more rigorous clinical data.

Burnout is resolved by changing what is causing the chronic stress, by rest, boundaries and, where needed, professional support.
